PROCEDURE, PRINCIPLE AND INTERPRETATION

PREPARATION OF BENEDICTS REAGENTS

  • Principle :
  • When the urine is boiled in alkaline copper sulphate, glucose and reducing substances in the urine reduces cupric ions to red cuprous ions.
  • The degree of column change corresponds to the concentration of reducing substances in the urine

PREPARATION OFREAGENT

  • Copper sulphate - 173grms
  • Tri sodium citrate - 173grms
  • Sodium bicarbonate -100grms
  • Distilled water 100ml

PROCEDURE

  • Dissolve the Tri sodium citrate and sodium bicarbonate in about 500ml distilled water by little warming.
  • Dissolve copper sulphate in another 500ml. After 2 mints cooling

INTERPRETATION

  • This benedicts solution will give false result to glucose, galactose, lactose, fructose and ascorbic also gives positive result

KOVACS REAGENT PREPARATION

PREPARATION OF KOVACS REAGENT

  • Chemicals :
  • iOS amyl or iOS butyl alcohol 150ml
  • Concentrated HCL 50ml
  • Para dimethyl amino benzaldehyde 10grms

PROCEDURE

  • Dissolve para dimethyl amine in the alcohol and slowly add the acid while constantly stirring the mixture
  • Reagent should be pale coloured and stored in brown bottle in refrigerator on turning brown discard the reagent the aldehyde may require gentle heating in order to go in to the solution.
